Captain Gaikwad struck 67 while Kartik hit 54 as Chennai reached their target of 160 with 11 balls to spare.
Seamer Anshul Kamboj starred with the ball for Chennai, removing opener Will Jacks for one in the second over and then returning to dismiss Hardik Pandya and Robin Minz on his way to figures of 3-32.
Naman Dhir top-scored for Mumbai with 57, putting on partnerships of 58 and 40 with Ryan Rickelton (37) and Suryakumar Yadav (21).
The latter's dismissal in the 11th over, leaving his side 99-3, stalled the Mumbai innings and they scored just 60 off their remaining 56 balls to close on 159-7 having chosen to bat.
Jasprit Bumrah had Sanju Samson caught behind for 11 in the second over and AM Ghazanfar bowled Urvil Patel for 24 in the sixth, but Chennai never really looked troubled.
The win moves the home side up to sixth, above Delhi Capitals on net run-rate, while Mumbai remain ninth after their third consecutive defeat.
